Improving evaluation of potato resistance to Rhizoctonia solani infection by optimizing inoculum-based method combined with toxin-based assay

چکیده

Rhizoctonia solani causes stem canker and significantly impacts potato production. Conventional assay to evaluate resistance using R. inoculum is time consuming. To establish an effective fast assay, 20 cultivars/lines were examined both AG2-1 as its derived toxin (RS toxin). In field trials of 2009 2010, wheat-bran-mediated was incorporated at either 0, 2, 3, 4 or 5 g per seed piece in the soil before sowing furrow. Stem evaluated sixty-five days after planting. Inoculation well distinguished cultivars/lines. For a quick screening, toxin-based method established by treating seedlings with RS AG2-1. prepared heating culture. Potato obtained through tissue culture grown Murashige Skoog medium. Seedlings stage 12 cm height transferred into agar amended incubated for eight days. The level inhibition caused positively correlated concentration. By evaluating various cultivars/liens that have different sensitivities toxin, from 33% 100% on stems sections 32% 148% seedlings. evaluation highly data pathogen (r = 0.731, P < 0.01). wheat bran-mediated 3 evaluations. improved efficiency disease evaluation. Both assays showed none completely resistant However, cultivar ‘Desiree’ had lowest disease, whereas ‘Atlantic’, ‘Favorita’, ‘Shepody’ most susceptible cultivars.

A simple method based on laboratory inoculum and field inoculum for evaluating potato resistance to black scurf caused by Rhizoctonia solani

A two-step method was developed to evaluate potato resistance to black scurf caused by Rhizoctonia solani. Tuber piece inoculum was first conducted in the laboratory, which was also first reported in this study. After inoculation with pathogen discs and culture for 48 h, the necrotic spots on the inoculated potato pieces were generated and measured by the crossing method. Further evaluation was...

Transformation of Potato (Solanum tuberosumcv.Savalan) by Chitinase and β-1,3-Glucanase Genes of Myco-Parasitic Fungi Towards Improving Resistance to Rhizoctonia solani AG-3

Potato (Solanum tuberosum L.) an agro-economically important food crop in the world, is sensitive to many fungal pathogens including Rhizoctonia solani (AG-3), the causal agent of stem and root rot diseases. Chitinase and glucanase are cell wall degrading enzymes which have been shown to have high antifungal activity against a wide range of phytopathogenic fungi. In the present study, plasmid p...

Evaluation of yield potential and resistance to rhizoctonia (Rhizoctonia solani) disease of new sugar beet (Beta vulgaris L.) hybrids

Rhizoctonia root rot disease is considered as one of the important diseases in sugar beet. Using resistant cultivars decreses production costs, especially application of fungicides and increases the efficiency in controlling of root rot disease. Therefore, growing of resistant cultivars is considered the best approach for disease control. To achieve this goal and to determine yield potential of...

The keto sugar, L-sorbose, inhibits hyphal extension and accumulate in the soil. When applied to seed in crystalline promotes profuse branching in Rhizoctonia solani. When form with sticker, L-sorbose is highly water soluble and may applied to cotton s...
